Why? 'Stem cell therapy is not a black and white type of event. There are varying degrees of success based on many factors of which the capacity of the cells is one. The functional capacity of a 44 year old cell is not as good as that of a 6 year old but that does not mean that they have no value. Our requirements for storing material mean that the cells demonstrate acceptable growth rates, expected cellular morphology and growth characteristics which indicate their ability to differentiate into tissue specific lineage cell types. If the cells do not meet our minimum criteria for usefulness we will not store them.

Aside from these tests we cannot make any specific guarantees about the cells usefulness. Adults should not be put off attempting to store their stem cells and can have faith that if we successfully complete the process of stem cell extraction then the cells are of high enough quality to be useful should they be required.

BioEden do not make any charge for the process of harvesting stem cells where no viable stem cells can be found.

It’s not exactly the elusive “Fountain of Youth” but it’s not far from it – this new method can restore muscle function and strength to the elderly actually matching a young, healthy state.

With the ever increasing population of elderly worldwide, loss of skeletal muscle function and strength has become a significant chronic condition. It is estimated that people lose 15% of muscle mass each year after the age of 75, and up until now that loss has been irreversible and often devastating. Skeletal muscles support basic functions such as sitting, standing, blinking, and swallowing. The loss of muscle function is not easy to deal with – and many of you know exactly what I’m talking about.

Co-author of the study Assistant Professor Penney Gilbert at University of Toronto notes that during aging some stem cells experience a protein modification that impedes the growth of new stem cells. According to Gilbert, “The new method allowed the aged cells to grow and make more copies of themselves.” By transplanting these rejuvenated stem cells into injured or aged muscles tissue, they were able to return strength to levels on par with young, healthy tissue.

Researchers have created an adult stem cell based method for restoring strength to damaged skeletal muscles in the elderly. 

Skeletal muscles are some of the most important muscles in the body, supporting functions such as sitting, standing, blinking and swallowing. In aging individuals, the function of these muscles significantly decreases.   People lose fifteen percent of muscle mass every single year after the age of 75, a trend that is irreversible.

Through tracing the signaling pathways of the cells, the researchers determined that during aging, a subpopulation of stem cells begin to express a modification of a protein that inhibits their ability to grow and make new stem cells.

"But if we instead treated those cells outside the body with a drug that prevented that protein modification from occurring, in combination with culturing the cells on something soft that is reminiscent of soft skeletal tissue, like a hydrogel biomaterial, the combination allowed the aged cells to grow and make more copies of themselves," says co-author and University of Toronto assistant professor Penney Gilbert. 

The rejuvenated cell cultures were then transplanted into injured and aged tissues, with remarkable results: the transplanted cells returned strength to the damaged and aged tissues to levels matching a young, healthy state.

Rather than trying to 'turn back the clock' on dysfunctional stem cells in the aged population, it stimulates stem cells from old muscle tissues that are still functional - to begin dividing and self-renewing.

One of the significant challenges to elderly individuals who receive hip transplants, for instance, is the challenge of repairing skeletal muscles around the hip joint injured during surgery. The study points to the potential for future post-surgery therapies that could leave elderly hip replacement patients spry in a fraction of the time.

"It's a really new, exciting field," says Gilbert, who argues that the muscle stem cell field, which only began to isolate muscle stem cells for study within the last five years, is especially "wide open" in Toronto where "there are really impassioned clinician researchers who are interested in restoring strength in aging and disease.

Though it was unintentional, she embarrassed Yamanaka, the winner of the 2012 Nobel Prize.

It’s not surprising that last week Haruko Obokata issued a plea for privacy. On Jan. 29 she published a scientific paper on stem cells that could revolutionize medicine, and overnight the researcher based at the Riken Center for Developmental Biology (CDB) in Kobe became a domestic and international star.

The onslaught of media attention went beyond what Obokata had discovered — a way to revert ordinary cells from the body into kansaibō (stem cells), cells with the ability to develop into any cell type. These cells, known as pluripotent stem cells, have long been seen as the key to regenerative medicine, because in theory they can be grown into new organs and tissues.
 
As a young female scientist, Obokata is something of a rarity in Japan, and many reports included details of her life it’s hard to imagine being discussed if she was a man. Her laboratory is painted pink and yellow, and is decorated with pictures of Moomin characters from children’s books.

She is 30, has a pet turtle — and a boyfriend. Instead of a lab coat, she wears a Japanese apron, a kappogi, originally designed to be worn over a kimono. The Internet shopping service Rakuten Ichiba reported a sharp increase in kappogi sales after the publicity.
But none of this would have been news had Obokata’s work not been so amazing. When we heard of it in the New Scientist office here in London, where I am the news editor, we just couldn’t believe it.

It is certainly possible to make pluripotent stem cells, and I’ve reported here about the work of another Japanese pioneer in that field, Shinya Yamanaka of Kyoto University. But Obokata’s method was so incredible in its simplicity that it almost sounded too good to be true.
Yamanaka had identified four genetic factors that, when transferred into an adult cell using a virus, could cause it to revert to a pluripotent stem cell. This breakthrough work won Yamanaka a share of a Nobel Prize — but one of the inherent problems there is that transferring the four genetic factors can also cause cancer.  

NOTE: Don Margolis has declared the 2012 Nobel Medicine prize was a fake, bribed by Big Pharma.  Yamanaka had then, and to this day, accomplished NOTHING beneficial to mankind.  Why did they do it? Because Pharma's embryonic stem cell hoax has been exposed as pure garbage, they had to come up with another reason for thr misinformed millions to avoid Adult Stem Cells.   These are the only stem cells which actually help people and therefore can potentially reduce Pharma's quarter-billion-a-day profits.

Obokata’s work seems to avoid cancer completely. 
 
Rather than using a virus and infecting cells with genetic factors, she simply stresses them. Squeezing cells through narrow tubes or bathing them in an acidic solution turns them pluripotent.
 
Surely, we thought, it can’t be so easy?
 
It turns out that the editors at Nature, the British journal where Obokata’s work was published, couldn’t believe it either. When she and her colleagues submitted the work for publication in 2012, it was rejected. She went away, worked on more experiments and resubmitted. This time, after yet more extensive checks, the journal accepted the paper. And the scientific — and general — media deluge began.

Other labs are already trying to replicate the work, and some scientists remain skeptical despite the extra-stringent review process prior to publication.

One of the first things I wondered was whether, if this is the real thing, Obokata and her team have patented the discovery. The secret to a simple method for making pluripotent stem cells could be worth a fortune. At a press conference just before publication, where she was asked about this, Obokata replied: “Yes, we have a patent” — before abruptly stopping. “Actually, I have to say ‘no comment,’ ” she said. Clearly there are plans in motion behind the scenes. The Asahi Shimbun then reported that Obokata’s group had started with patent applications in April 2013.

We can expect big things. One of the reasons this has caused such a sensation is that it comes out of left field. Her work is a great advert for thinking and working differently from the crowd — not a traditional approach for a Japanese scientist. Obokata’s co-author, Yoshiki Sasai, also at the Riken CDB, praised the approach, saying, “She held her own view backed by data.”
 
Riken CDB is abuzz with important work at the moment. As I reported here in December, another stem-cell biologist there, Masayo Takahashi, is about to conduct the world’s first stem-cell trial on humans. In an interview with New Scientist, she praised Obokata’s discovery, saying, “I feel joyful that we are both working in this wonderful area of science.”
 
So it’s a fascinating time — and fantastic to see women pioneering this research. However, Japan’s media still has a long way to go regarding its portrayal of women. After all, a piece in the Yomiuri Shimbun soon after Obokata’s paper was published ended with the cheesy observation that “she always strives to be fashionable as well.”

Final Note:  RSCI applauds this amazing young woman and hopes that she succeeds in proving the validity of her work over time.  However, RSCI also knows that given the structure of all medical systems in the entire developed world, there is no chance of anyone being treated with this method (outside clinical trials) before 2030.

Occasionally we come across technology that’s not really technology in its truest sense. Sometimes, technology is really an assistant to nature. This is particularly evident in certain functions of the human body.

The terms ‘regrow’ and ‘regenerate’ often make people think of regrown limbs. Agreed, that’s not entirely possible…for now. But the body can regenerate thanks to ‘wonder-cells’ known as stem cells.
 
Although stem cells are naturally occurring, the latest technology is maximising their potential. Thanks to scientists and researchers we can do more now with stem cells than ever before.


It All Just Works Like Magic!
 
Stem cells can repair and regrow vital systems in our body. And encouragingly stem cells occur naturally in all of us. In fact the reason we are who we are is because while we’re embryos stem cells chop and change about to make all the bits that make us human.
 
Without getting too bogged down in the biology and science, there are effectively two types of stem cells. The first kind is embryonic stem cells, also known as pluripotent stem cells. These have the ability to become every type of cell in the body.
 
The second kind is adult stem cells, also known as multipotent stem cells. These have the ability to become only certain types of cells in the body.
 
These wonder-cells are vital to the future of regenerative medicine. That is the practice of medicine that repairs and regenerates the body. The potential lies in the ability to one day rewind the entire process of aging.
 
Stem cells are so important for a few key reasons. The Australian Stem Cell Foundation outlines these on their website,

‘Stem cells are different from other cells in the body in three main ways:
 
‘1. Stem cells are unspecialised. They have not developed into cells that perform a specific function.
 
‘2. Stem cells can differentiate. This means they can divide and produce cells that have the potential to become other more specific cell types, tissues or organs. These new cells and tissues are used to repair or replace damaged or diseased cells in the body. Once cells have differentiated, they have less capacity to form multiple different cell types, and become ‘committed’ to becoming a particular cell type. Skin stem cells, for example, give rise to new skin cells when needed, to assist regeneration after damage and as part of the normal ageing process.
 
‘3. Stem cells are capable of self-renewal. Stem cells are able to divide and produce copies of themselves which leads to self-renewal. Once a cell has become specialised (has differentiated) to a particular tissue or organ, it has a very limited capacity to self-renew (produce new stem cells) but instead produces only cells relevant to that organ.‘

Medical technology means scientists and researchers can use stems cells in a number of ways to cure disease and ailments like what I’m about to explain below. In what seems like a magical trick, scientists have even brought ‘dead’ organs back to life using stem cells.
 
And there’s growing evidence that stem cells hold even more potential than first thought.


The Human Element of These Amazing Cells
 
David Pyne is 60 years old. He lives in Manchester, England. He has four children and is a Grandfather. David got the worst news of his life in August 2012. He had leukaemia. He had about 12 to 18 months to live.
 
One way to treat leukaemia is to get a bone marrow transplant. However a global search resulted in no match for the required transplant. Time was running out and so were his options to stay alive.
Thankfully two babies were born. One in America, one in France. And even better was both mothers had decided to donate their umbilical cord blood.
 
This was so significant because one of the richest sources of stem cells is umbilical cord blood. It also just happened that these two donors were a good match for David.
 
With no other option the doctors treated him with the new stem cells. He spent six weeks in hospital hoping this would be the solution he so desperately needed.
 
As reported in The Mirror, the director of the hospital where David was treated said,

‘Umbilical cord blood is very rich in stem cells, which being so immature have phenomenal regenerative powers. These were a great alternative source of cells for David, in fact the only option, as after a worldwide search he had no other donor.‘

The stem cells worked, and David is now in remission. The regenerative powers of the stem cells have literally saved his life.
 
With more and more research every year, there’s seemingly endless potential in what stem cells are capable of treating. And in some cases stem cells might even improve parts of the body better than they were before.

Umbilical cord blood stem cells have become an extremely valuable alternative to bone marrow adult stem cell transplants, ever since cord blood stem cells were first used for patients over 25 years ago. The first umbilical cord blood stem cell transplant was performed in October 1988, for a 5-year-old child with Fanconi anemia, a serious condition where the bone marrow fails to make blood cells. That patient is currently alive and healthy, 25 years after the cord blood stem cell transplant.

Since that time, over 30,000 cord blood stem cell transplants have been done around the world, and transplants have increased for various blood and bone marrow diseases and leukemias, as well as for genetic enzymatic diseases in children. Cord blood stem cell transplants have also become more common for adults with leukemia. Cord blood transplants have been especially helpful for racial and ethnic minorities.
 
Bone marrow adult stem cell transplants require an exact match between donor and recipient, and it can sometimes be difficult to find a donor match for a patient, especially for minorities. But umbilical cord blood stem cells can be used with some mismatch and still provide successful treatments.**
 
The Wall Street Journal recently noted the increased interest in umbilical cord blood by scientists and doctors seeking stem cell cures. Besides current treatments, cord blood stem cells are now being studied for their potential to treat many more diseases, including Type 1 diabetes and rheumatoid arthritis, as well as congenital heart disease and cerebral palsy. The story quotes Dr. William Shearer, professor of pediatrics and immunology at Baylor College of Medicine:
 
“It’s a disposable item that Mother Nature provides us with… It’s a renewable source. It’s free and why not use it?”
Since the first umbilical cord blood stem cell transplant over 25 years ago, over 600,000 cord blood units have been stored away around the globe for future lifesaving transplants. Just two examples of public programs to collect and store umbilical cord blood stem cells are the National Marrow Donor Program (motto: “You could cure someone’s blood cancer by giving birth”) and the National Cord Blood Program, and additionally there are commercial cord blood storage companies, involved in collection, storage, and research. The data so far show that cord blood stem cells can be stored frozen for over 20 years without loss of potency.
 
And it’s not controversial. As a recent news story in the Washington Times showed, many more states are turning to ethical, successful adult stem cells, providing real hope and real treatments for thousands of people. One such state, Kansas, last year initiated a unique Midwest Stem Cell Therapy Center that will treat patients, do research on new therapies, educate the public and professionals on the advantages of adult stem cells such as those from cord blood and the solid umbilical cord, and train physicians to deliver those treatments. Paul Wagle was appointed by Governor Brownback to represent the patient community on the new Advisory Board for the Kansas Center. Paul received an umbilical cord blood stem cell transplant for his leukemia in 2005. Partly as a result of the successful treatment, Paul developed an interest in science and earned a triple major from Benedictine College in Kansas in 2013, and is now in seminary. The Kansas Center has already treated its first patient and held its inaugural scientific conference.

Here are just a few other examples of the double lifesaving from a born baby and the saved cord blood.

Mary Lou Rusco also received umbilical cord blood stem cells for her leukemia. She received the treatment from doctors at the Kansas University Medical Center, and is now free from leukemia.

Joe Davis, Jr. was diagnosed with sickle cell anemia, at only a few months old. His parents were told that he wouldn’t survive to be a teenager, and they couldn’t find a bone marrow match for him. But along came younger brother Isaac, whose umbilical cord blood stem cells saved Joe Junior’s life.

Chloe Levine received an innovative cord blood stem cell transplant at Duke University to treat her cerebral palsy. She’s now a happy healthy little girl.

**As accurate and meaningful as this excerpt is, it is too conservative in 2014.  Yes, in 2010 it may have been considered correct, but today, umbilical cord stem cells (UCSC) are rapidly becoming the adult stem cells of choice at our world- leading 14 clinics.  There are two reasons for this: (1) Matching UCSC, as they must for bone marrow transplants, is a waste of time. The ultra-modern, super-clean cutting-edge labs which produce them deliver a product without DNA markers.  This means that the receiving patient's immune system cannot reject them as "strange DNA."  (2) Even more exciting, is that the stem cell leaders are choosing the rare "mesenchymal" stem cells rather than the commonplace stem cells found in bone marrow, adipose, or umbilical cords because they can become virtually any kind of tissue cell the patient may need.  Since the mesenchymal numbers are something like 1/1000 of the common cells, they must be cultured and grown over time---normally 2-3 months with bone marrow and adipose, making the patient wait or make two trips to the clinic.  But the new M-UCSC are prepared and delivered to the treating doctors before the patient even arrives, meaning he now gets mesenchymals without any delay!---Repair Stem Cells Institute

A man who was given 18 months to live after being diagnosed with leukaemia has said his life has been saved by stem cells taken from umbilical cords.

David Pyne, from Baguley, Manchester, was given a transplant of cells donated following births in France and the US.
 
The 60-year-old, who is in remission, was forced to consider alternative treatments after chemotherapy failed.
 
He said being told there was a chance that newborns could save his life was "incredible news".
 
The treatment for patients with cancers such as leukaemia uses donated blood stem cells, usually from adult donors, to replace damaged ones.


'Regenerate bone marrow'

No suitable matches were found for Mr Pyne through a search of his family and a database of other donors.
 
The grandfather, who underwent the transplant at Manchester's Christie Hospital, said he "had never heard of getting stem cells from umbilical cords".
 
"To hear that there was a chance that newborns could save my life was incredible news".
 
In the UK, pregnant mothers are given the option to donate and the use of stem cells to treat cancer is available on the NHS. The Christie has carried out six transplants over the past year.
 
The hospital's Dr Mike Dennis said the treatment was a "variant of a blood transfusion".
 
"The cord blood has been frozen anywhere in the world and it can be flown to where the patient is being treated," he said.
 
"It can then be given to them after the chemotherapy and radiotherapy as a life-saving procedure to regenerate their entire bone marrow."
 
Since the transplant, Mr Pyne has been treated as an outpatient at the hospital, attending weekly check-ups.

For more than a decade, Roberto Bolli, MD, has been working to revolutionize the treatment of heart failure by providing patients treatment derived from their own cardiac stem cells. Today, the University of Louisville announced that the Jewish Heritage Fund for Excellence is providing just over $2 million to help Bolli move his research another step closer to being able to help potentially millions of people throughout the world.

The university also announced it was providing nearly $1 million from proceeds from a previous gift from former UofL board chair the late Owsley Frazier, bringing the total to $3 million in support of Bolli’s groundbreaking work.

The funds will be used to purchase and install a Current Good Manufacturing Practices facility required by the Food and Drug Administration for processing the stem cells for use in the study participants.

“Through nearly five years of research studies Dr. Bolli has provided patients who have had heart attacks infusions of their own cardiac stem cells and finding that their hearts are actually regenerating heart muscle severely damaged by heart attack,” said James Ramsey, Ph.D., president of the University of Louisville. “This gift from the Jewish Heritage Fund for Excellence will enable Dr. Bolli and his team to significantly expand his clinical trials with the goal of transforming the lives of hundreds of people, and eventually millions.”

Bolli and his team, which includes Dr. Piero Anversa at Brigham and Women’s Hospital/Harvard Medical School in Boston, harvest cardiac stem cells, referred to as “c-kit positive” cells because they express the c-kit protein on their surface, from the patients during coronary artery bypass surgery conducted at Jewish Hospital in Louisville. The stem cells currently are purified in Anversa’s lab in Boston and allowed to grow. Once an adequate number of stem cells are produced – about one million – Bolli’s team in Louisville reintroduces them into the region of the patient’s heart that has been destroyed by cardiac infarction, otherwise known as heart attack. The new facility will allow the purification and growing processes to be performed in Louisville.

“As part of our mission, the Jewish Heritage Fund for Excellence provides support for innovative medical research that will significantly impact the lives of people in our community and beyond,” said Louis Waterman, chair of the board. “Dr. Bolli’s work has the potential to change how heart failure is treated. It is our privilege to facilitate his, and his team’s, efforts through this gift.”

“Dr. Bolli is recognized internationally as an innovative leader in the treatment of heart disease and heart failure,” said David L. Dunn, MD, PhD, UofL executive vice president for health affairs. “His work has the potential to forever alter how we view heart failure and how we provide care for the patients who suffer from it.”

Bolli, professor of medicine and the director of the Institute of Molecular Cardiology at UofL, recently was honored with the American Heart Association’s Research Achievement Award for his work with cardiovascular research. He also is editor-in-chief of the journal Circulation Research.

“These resources will enable our team to significantly enhance our efforts to bring a new way of treating heart failure to patients who so desperately need help,” said Bolli, who also holds the Jewish Hospital Heart & Lung Institute Distinguished Chair in Cardiology. “In our initial trials we have demonstrated that we are able to reverse the damage caused by a heart attack. I am very excited that we now will be able to explore this further in a much larger population.”via
